Bone morphogenetic protein 2 ( BMP-2) is one of at least 15 structurally and functionally related BMPs, which are members of the transforming growth factor β (TGF- β) superfamily of cytokines. BMPs are identified as protein regulators of bone and cartilage formation. Similary to other TGF- β family proteins, BMPs are highly conserved across animal species.
The functional form of recombinant human BMP-2 is a 26kDa protein composed of two identical 115 amino acid polypeptide chains (monomers) linked by a single disulfide bond.
BMPs initiate their signaling cascade by binding to a dimeric complex of two transmembrane serine–threonine kinase receptors, termed type I and type II. The activated receptor kinases subsequently phosphorylate transcriptional factors, called Smads, which activate the expression of target genes in concert with coactivators.
BMPs are of tremendous interest as therapeutic agents
for healing bone fractures, preventing osteoporosis, treating periodontal bone defects and enhancing bone response around alloplastic materials implanted in bone.

Formulation: rhBMP-2 was lyophilized from a 0.2 µm filtered solution after extensive dialysis against 50mM acetic acid.
Reconstitution: It is recommended to reconstitute the lyophilized rhBMP-2 in sterile 50mM acetic acid not less than 100 µg/mL.
Protein content: Protein quantitation was measured by UV spectroscopy at 280 nm using the absorbency value.
Purity: > 95% as determined by SDS-PAGE and visualized by Coomassie stain.
Biological Activity: Measured by its ability to induce alkaline phosphatase production by mouse C2C12 mioblastic cells. The ED50 for this effect is 1-2 µg/mL
Storage: Lyophilized rhBMP-2 should be stored at -20 °C although it is stable at room temperature for a few weeks. Upon reconstitution may be stored at 4ºC for short periods. For long-term storage, freeze in working aliquots. Avoid repeated freeze-thaw cycles.
